Abstract
Objectives: The search for new serum markers related to aging, atherosclerosis and predictors of cardio-vascular events is important in modern society, especially in regions and cities with high mortality from cardiovascular diseases. Study design: Activity of serum matrix metalloproteases (MM Ps), chitotriosidase and cystatin C concentration were assessed in healthy persons (n=26, aged 20-45 years), and elderly statin treated (n=25, aged 50-65 years) and non-treated (n=25, aged 50-65 years) persons, and a group of patients with atherosclerosis (n=32) that were about to undergo a cardiovascular surgical operation. Methods: Activity of MMP and chitotriosidase was measured by fluorescent methods against M CA-Pro-Leu-G ly-Leu-DpA-Ala-Arg-N H, and 4-MUF-~-D-N,N ',N " triacetylchitotrioside as substrates. In addition, hs-CRP and cystatin C concentration were determined. Results: Increased serum MMP and chitotriosidase activity, cystatin C concentration in elderly group, as well as increased hs-CRP concentration were demonstrated. Similar results were obtained in patients with atherosclerosis before the coronary shunting operation. However, MMP and chitotriosidase activity as well as hs-CRP were elevated more considerably compared with elderly persons of the same age. Statin treatment lowered hs-CRP concentration and MMP activity in elderly group. Conclusions: Increased serum MMP, chitotriosidase activity and cystatin C concentration are non-lipid markers of aging and early atherosclerosis development. MMP activity appears to be one of the possible markers of clinically diagnosed atherosclerosis and indicates efficacy of lipid-lowering statin therapy in elderly people.
